Reading the Iodine Test
Core point
A test you have never calibrated is an opinion with a dropper. Before you trust your read on a real mash, run the test once on a sample you know contains starch and once on one you know does not — then you know what your iodine, your light, and your eyes actually show you.
What the test is
Iodine solution reacts with starch — the long, unconverted glucose chains — and the complex it forms reads as a deep blue to blue-black stain. Short fragments and fermentable sugars do not produce it. So the test is a starch detector, nothing more: it tells you whether long chains remain, and it says nothing about which fermentable sugars conversion produced — that question belongs to The Sugar Bible.
In this site's process the test has one job at two moments: confirming that liquefaction and conversion actually finished, on the way to a final wort that is iodine negative.
Calibrate your eye first
Run the test on two knowns before you ever rely on it in a real brew day. This costs five minutes once.
- A guaranteed positive. A pinch of flour or crushed raw sorghum stirred into warm water. Drop iodine on a spoonful. The stain you see — dark blue to blue-black, unmistakable — is what "starch present" looks like with your iodine, your lighting, and your sample size. In Bard's production records that is exactly how a failed conversion announced itself: the mash sample "turned iodine blue, loaded with starch."
- A guaranteed negative. Plain water, same iodine, same light. What you see is the iodine's own amber/tea color, unchanged. That — not some imagined pure white — is what "negative" looks like.
Everything you will ever read from this test is one of those two references or something in between. An in-between, murky read on a real sample is information too: treat it as "not clearly negative" and act accordingly, rather than rounding it in the direction you were hoping for.
Running it on a real mash
- Pull a small sample onto a white surface — a white plate or dish makes the color readable in brewhouse light.
- Let it cool for a moment; drop the iodine beside or into it and read promptly.
- Record the read with its context: sample point, time, temperature, result, and whether the sample was mash or wort. A result without its sample point cannot be interpreted later — and later is when you will want it.
The interpretation traps
These are where the test goes wrong in practice — the chemistry rarely does.
An intermediate positive is not the final result. A mash sample can read positive at the end of a hold while the batch is still on track: starch-degrading enzymes keep working during wort separation, so the number that matters is the final wort, which should be iodine negative. Do not extend holds or redose on the strength of one mid-process positive — Mash Protocol 1 carries this rule at the point of action.
A grain fleck is not a mash read. Iodine dropped on an intact particle of grain will find the starch inside it and stain dark, even in a well-converted mash. Read the liquid, and if visible starch on grain plus a positive liquid read appear together, believe the combination — that is the practical warning that conversion is incomplete (Sorghum Mash Challenges owns what to do next).
Negative does not mean "good wort." Iodine-negative means the long chains are gone. It does not tell you the fermentability profile, the gravity, or whether the wort will ferment to plan — those are separate measurements. The test clears one failure mode only.
A persistent positive is a process signal, not a dosing signal. In the archived production record, the memorable failure read blue after an hour, read blue again after more enzyme and another hour, and the batch was dumped — because the problem was upstream of the enzyme addition. A positive that will not clear means find the bottleneck — starch access, temperature, sequence — not add more (find the actual bottleneck).
Limits of this page
This page teaches the bench read as this site's production experience and records support it. It publishes no iodine product, concentration, or dose — use a standard brewing iodine/iodophor test solution per its own directions, and keep it out of the beer: the test sample is waste, never returned to the kettle. Photographic color references are a planned addition; until then, the two-knowns calibration above is the anchor, and it is the more transferable one — it calibrates your materials, not ours.
